A Phase II Study of GW572016 and Tamoxifen in Patients with Metastatic Breast Cancer Resistant to Single Agent Tamoxifen |

| Description: |
The purpose of this study is to determine how safe and effective GW572016 in combination with tamoxifen will be for the treatment of advanced breast cancer. Patients will take six pills of GW572016 and one pill of tamoxifen, amounting to seven pills daily as well as keep a diary. |
| Criteria: |
- Age ≥ 18 years
- Primary adenocarcinoma of the breast confirmed by histology or cytology. ER and/or PR+
- Locally advanced or metastatic disease not amenable to surgery or radiation with curative intent. Must not have rapidly progressive disease
- Must have failed hormonal manipulation with tamoxifen. May have received treatment with an aromatase inhibitor as first line therapy for metastatic disease prior to treatment with tamoxifen, but no other hormonal manipulation upon failing tamoxifen
- ECOG performance status 0 – 2 6. Patients must have normal bone marrow, liver and kidney function.
